Crowds gathered at the Libyan leader's residential compound chanting "we want revenge for our victims".
Colonel Gaddafi's 29-year-old son is believed to have died after a missile fired from a Nato war plane hit a house in a wealthy residential area of the capital, Tripoli.
Three of Gaddafi’s grandsons were also killed, and other friends and relatives injured.
The Libyan leader himself was also in the large one-storey villa but was not injured.
